The commercial foundation of RNA drugs predates COVID-19 vaccines. In 1998, the first oligonucleotide drug Fomivirsen was approved; in 2016, Spinraza demonstrated that antisense oligonucleotides could alter SMN2 splicing and improve motor milestones and survival in spinal muscular atrophy patients; in 2018, Onpattro validated the pathway of lipid nanoparticle delivery of siRNA to silence the liver TTR gene.

The capital path has shifted from single drug development to platform integration. Alnylam has established a product line around RNAi, GalNAc conjugation, and liver targeting; Moderna has built a manufacturing system from vaccines to tumor therapies around mRNA design and lipid nanoparticles; Vertex and CRISPR Therapeutics' Casgevy was approved in 2023, showing that RNA-guided gene editing can also enter clinical commercialization. The truly scarce resources are not nucleotide synthesis, but delivery, toxicology, CMC production, clinical development, and payment access capabilities.

Industry positioning can be compared to the evolution of monoclonal antibodies: initially, the technological value came from "can specific molecules be manufactured," and then competition shifted to target combinations, production scale, and indication expansion. RNA has currently crossed the validation period for a few rare diseases and is entering a broad indication expansion period for cardiovascular diseases, tumors, and chronic diseases; however, solid tissue delivery remains an engineering challenge that determines platform boundaries, similar to early antibodies penetrating tumor microenvironments.

The essence is technological substitution: drug development is shifting from finding small molecules that can accidentally bind to targets to directly regulating protein production and gene expression through "programmable" interventions. The change is happening because sequencing, RNA chemical modifications, delivery materials, and computational design have simultaneously reduced the time cost from target discovery to candidate molecule generation; however, clinical efficacy is still determined by biological delivery and human safety, and code-based design cannot bypass human trials.
